Fig. 2. 15-deoxy-Δ12,14-prostaglandin J2 (15d-PGJ2)-induced apoptosis of breast cancer cells. (A) Differential P-STAT3Y705 expression in breast cancer cell lines. The basal levels of P-STAT3Y705 in MCF10A-ras, MCF-7, MDA-MB-231 and MDA-MB-468 cells as well as a non-oncogenic human breast epithelial MCF10A cell line were assessed by Western blot analysis. (B) MCF10A, MCF10A-ras, MCF-7, MDA-MB-231, and MDA-MB-468 cells were exposed to 15d-PGJ2 (10 or 30 μM) for 24 hours. The cellular viabilities were determined by the MTT assay. The values represent mean ± SD of the three independent experiments. DMSO, Dimethyl sulfoxide. **P < 0.01; ***P < 0.001 versus the corresponding control. (C) 15d-PGJ2 induces apoptosis in human breast cancer cell lines. Western blot analysis was performed with lysates from MDA-MB-231, MDA-MB-468 and MCF-7 cells after 24-hour treatment with 15d-PGJ2.
