Fig. 4. Effect of DHA on TNF-α- or poly (I:C)-induced disruption of mitochondrial membrane potential (MMP) in pancreatic stellate cells (PSCs). MMP was determined by confocal microscopy of cells pre-treated with DHA (50 μM) for 1 hour and stimulated with TNF-α (10 ng/mL) or poly (I:C) (1 μg/mL) for 1 hour prior to staining with JC-1 fluorescent dye (×200 magnification). (A) High MMP is indicated by red fluorescence and low MMP by green fluorescence. (B) The ratio of the intensities of red to green fluorescence was determined for the images shown in the (A). Values are expressed as the mean ± SE. DHA, docosahexaenoic acid; poly (I:C), polyinosinic-polycytidylic acid. *P < 0.05 vs. untreated cells; P < 0.05 vs. cells treated with TNF-α or poly (I:C) alone.
