Fig. 3. Effect of withaferin-A on interleukin-6 (IL-6)-induced STAT3 transcriptional activation. (A) STAT3 transcriptional activity assay using the nuclear extract from HCT116 cells treated with the indicated concentrations of withaferin-A with or without IL-6 (10 ng/mL) stimulation for 12 hours. Results are representative of three individual experiments. All data are presented as the means ± SE; t-test and ANOVA. **P < 0.01. (B) The nuclear translocation of STAT3 in HCT116 cells after treatment with IL-6 (10 ng/mL) was inhibited by withaferin-A. For immunofluorescence staining, cells were permeabilized and stained with anti-STAT-3 antibody after fixation (green). Anti-histone H4 antibody was used to stain the cell nuclei B (red).
