Fig. 5. Effects of genistein on GPR30 expression and reactive oxygen species (ROS) generation. (A) MDA-MB-231 and HCC1937 cells were treated with 50 μM Genistein. After 48 hours of incubation, the mRNA expression of GPR30 was assessed by reverse transcription-PCR. (B) The intracellular ROS levels in genistein treated MDA-MB-231 and HCC1937 cells were measured using a DCF-DA fluorescent dye. Cells were exposed to either dimethyl sulfoxide or 50 μM genistein for 24 hours. Images of cellular fluorescence were acquired by using a live cell image microscope. (C) The protein level of Nrf2 in HCC1937 with and without genistein treatment for 72 hours was measured by Western blot analysis. GAPDH, glyceraldehyde 3-phosphate dehydrogenase.
