Fig. 4. Involvement of genistein in the cell cycle arrest at the G2/M phase. (A) MDA-MB-231 cells were transfected with either scrambled siRNA or BRCA1-siRNA, and the effects of genistein on cell cycle progression were assessed. MDA-MB-231 cells were treated with 50 μM genistein for 72 hours. Harvested cells were washed using PBS and fixed with ice-cold 70% ethanol for 2 hours at 4°C. Fixed cells were washed with HBSS, followed by staining with propidium iodide. Cell population was measured by FACS analysis. (B) The experiment was repeated using MDA-MB-231 cells with and without BRCA1-siRNA knockdown to compare the effect of genistein on cyclin B1 expression levels. (C) The effects of genestein on cyclin B1 expression was compared in both MDA-MB-231 and HCC1937 cells. (D) MDA-MB-231 cells were transfected with either scrambled siRNA or BRCA1-siRNA, and then treated with genistein. Expression and phosphorylation levels of Akt were determined by Western blot analysis.
