Fig. 3. Inhibition of NF-κB signaling by baicalein (Bai). (A) Whole protein extracts were used for detection of inhibitor of κBα (IκBα) and phosphorylated p65. Cytoplasmic and nuclear extracts were used for detecting pIκBα and nuclear accumulation of p65. (B) IκB kinase β (IKKβ) was precipitated with an anti-IKKβ antibody, followed by a kinase assay using IκBα as the substrate. The relative band intensity was normalized by the immunoglobulin G (IgG). (C) Nuclear extracts were incubated with the [γ-32P]-labeled polynucleotide probe containing the consensus motif of NF-κB. A subsequent electrophoresis was performed to separate protein–probe complexes from free probe. The relative DNA binding activity was calculated.
